Abstract

Le récepteur B₁ des kinines est impliqué dans les phénomènes inflammatoires. Ce récepteur est inductible suivant différents types de dommages tissulaires et doit être soumis à une forte régulation. Le travail réalisé a permis de caractériser la régulation transcriptionnelle, traductionnelle et post-traductionnel. J'ai pu démontrer l'implication du facteur de transcription NF-κB dans l'induction du récepteur B₁ par les cytokines et le FBS. L'effet de la cycloheximide est partiellement régulé par ce facteur, mais elle joue certainement aussi un rôle (encore indéterminé) dans la stabilisation de l'ARNm. J'ai aussi démontré qu'il n'y avait pas de synthèse accrue du récepteur B₁ sous l'influence de son ligand dans les cellules de muscle lisse, les aortes fraîchement isolées ainsi qu'in vivo. Finalement, j'ai prouvé que le récepteur B₁ fluorescent est transloqué aux cavéoles suite à sa stimulation avec l'agoniste.

The B₁ kinin receptor is involved in inflammatory phenomena. This receptor is inducible according to different types of tissue damage and must be subjected to strong regulation. The work carried out has made it possible to characterize transcriptional, translational and post-translational regulation. I was able to demonstrate the involvement of the transcription factor NF-κB in the induction of the B₁ receptor by cytokines and FBS. The effect of cycloheximide is partially regulated by this factor, but it certainly also plays a role (as yet undetermined) in mRNA stabilization. I also demonstrated that there was no increased synthesis of the B₁ receptor under the influence of its ligand in smooth muscle cells, freshly isolated aortas as well as in vivo. Finally, I proved that the fluorescent B 1 receptor is translocated to the caveola following its stimulation with the agonist.
