Abstract

Les travaux contenus dans cette these portent sur la caracterisation des mecanismes de reparation de deux types de lesions de l'acide deoxyribonucleique (ADN), soit les sites apuriniques et apyrimidiniques (AP), soit les dommages crees par la bleomycine (BLM), en utilisant principalement la levure Saccharomyces cerevisiae comme modele. Dans un premier temps nous avons caracterise plus amplement le mode de reparation de l'ADN abasique par Apn1, l'AP endonuclease majeure de S. cerevisiae. Nous montrons qu'elle peut etre fonctionnellement remplacee par une AP endonuclease ou AP lyase bacterienne. Corroborant l'importance d'Apn1 dans la stabilite du genome de levure, nous presentons le clonage de genes homologues chez la levure S. pombe et le nematode Caenorhabditis elegans. En deuxieme lieu, nous explorons le role d'Imp2, un activateur de transcription, dans le phenomene de reparation de l'ADN endommage a la bleomycine ainsi que dans le controle des processus osmoregulatoires chez la levure Saccharomyces cerevisiae.

The work contained in this thesis focuses on the characterization of the repair mechanisms of two types of deoxyribonucleic acid (DNA) lesions, either apurinic and apyrimidine sites (AP), or damage created by bleomycin (BLM), in mainly using the yeast Saccharomyces cerevisiae as a model. Firstly, we more fully characterized the mode of abasic DNA repair by Apn1, the major AP endonuclease of S. cerevisiae. We show that it can be functionally replaced by a bacterial AP endonuclease or AP lyase. Corroborating the importance of Apn1 in the stability of the yeast genome, we present the cloning of homologous genes in the yeast S. pombe and the nematode Caenorhabditis elegans. Secondly, we explore the role of Imp2, a transcription activator, in the repair of damaged DNA with bleomycin as well as in the control of osmoregulatory processes in the yeast Saccharomyces cerevisiae.
